Title: Formulation and evaluation of fast disintegrating tablets using polyfunctional matrix
Introduction:
Coprocessed excipients are widely used in the manufacture of fast
disintegrating tablets by simple physical mixing followed by direct
compression. They have good rheological properties and make it possible to
correct the poor compressibility properties of these active principles. Due to
swallowing difficulties, rapidly disintegrating tablets are used for children
and the elderly. Method: The purpose of this work is to formulate an
orodispersible tablet by developing a fast disintegrating matrix that
represents a charge excipient for these tablets. The excipients used are:
mannitol as a diluent, corn starch as a binder, crospovidone as a
disintegrating, colloidal silica as a flow and permeabilizing agent and sodium
lauryl sulphate as a lubricant. For a first approach, preliminary tests made it
possible to define the proportion of the binder in the formula. Subsequently
the retained formula was improved by incorporating a disintegrating and a flow
agent. The matrix obtained meets the objectives initially set; disintegration
time less than 3 minutes and sufficient hardness of the tablets. This matrix
has been applied to several active substances. Results: pharmacotechnical
controls are carried out both on the granule and on the tablet (placebo),
residual moisture content of the granules is 3, 2%, flow time=7,76 ±0,35
seconds, friability=0,53%±0,02, disintegration time=30±2 seconds, wetting
time=28±0,81 seconds. Direct compression was used to prepare tablets contains
furosémide. The tablets obtained were evaluated: content uniformity=102, 51 %,
disintegration time=25 seconds, dissolution test=not less than 99% in 15
minutes.
The
results are in accordance with specifications of pharmacopeia.
